COMMENTTM0269 is predicted to form a complex with TM0268, containing three domains of Methionine synthase (also termed Methyltetrahydrofolate S-homocysteine methyltransferase (EC:2.1.1.13)) responsible for binding of: homocysteine, methyltetrahydrofolate and cobalamine cofactor.
TM0268 alone should be capable of performing the main reaction: conversion of homocysteine to methionine using methyltetrahydrofolate as a methyl donor via cobalamine cofactor. However it would lack the ability of reactivation (as needed to compensate for occasional oxidation of the cofactor). In human and E.coli MetH this function is performed by the C-terminal domain.
TM0269, a distant homolog of the missing C-terminal domain, is expected to compensate for this function.
Genome context: an operon (chromosomal cluster) containing genes encoding for TM0268/TM0269 and TM0270(methylenetetrahydrofolate reductase (EC 1.5.1.20), involved in generation of MetH substrate, methyltetrahydrofolate).
